Azithromycin tablets ip 500mg use to treatment for bacterial infection like bronchitis; pneumonia, sexually transmitted diseases (STD); and infections of the ears, lungs, sinuses, skin, throat, and reproductive organs. Class of drug: ( Macrolides )
Storage of azithromycin:
- Intravenous: Store below 30°C.
- Oral: Store below 30°C.
Mechanism of Action:
Azithromycin tablets ip 500mg blocks the transpeptidation by binding to 50s ribosomal subunit of susceptible organisms and disrupting RNA-dependent protein synthesis at the chain elongation step.
Absorption: Reduced by food (capsule formulation); peak plasma concentrations after 2-3 hour.
Distribution: Azithromycin extensive into the tissues (concentrations higher than those in blood), WBC (high concentrations), CSF (small amounts).
Metabolism of azithromycin : Liver (demethylation).
Azithromycin excretion: Via the bile (as unchanged drug and metabolites); via the urine (6% of the dose). Elimination and half-life: about 68 hr.
Indication & Dosage of azithromycin:
1. Oral use azithromycin tablets ip 500mg:
Azithromycin use for Skin and soft tissue infections and Respiratory tract infections:
- For adult: 500 mg once a daily for 3 days, and 250 mg once a daily for 4 days.
- For child: Less than 6 month 10 mg/kg. 15–25 kg person: 200 mg, 26–35 kg: 300 mg, 36–45 kg: 400 mg. Doses to be taken once daily for 3 days.
Uncomplicated genital infections due to Chlamydia trachomatis:
- For adult: 1 g as a single dose.
Uncomplicated gonorrhoea:
- Adult use : 2 g as a single dose.
Azithromycin use for Prophylaxis of disseminated Mycobacterium avium complex (MAC) infections:
- Adult: 1.2 g once every week for treatment or secondary prophylaxis: 500 mg once daily with other anti mycobacterial.
- Child: Less than 6 month: 10 mg/kg once daily for 3 days.
In case of Granuloma inguinale use azithromycin:
- For adult use: Initially, 1 g followed by 500 mg daily. Alternatively, 1g once a week for at least 3 week, until all lesions have
completely heal.
2. Intravenous use azithromycin:
Active immunigation against typhoid fever caused by Salmonella typhi:
- Adult use : 500 mg once daily for 7 days.
How to take azithromycin tablets ip 500mg ?
- Tablets: May be taken with or without food. And may be taken with meals to reduce GI discomfort.
- Suspension: May be taken with or without food. May be taken w/ meals to reduce GI discomfort.
- Capsule: Should be taken on an empty stomach. (Take on an empty stomach 1 hr before or 2 hr after meals.)
If miss the dose:
Do not take 2 tablet at a time you can start same dose.
Contraindications of azithromycin tablets ip 500mg :
Main contraindication is hypersensitivity in your body.
What is special precautions of azithromycin is?
If some person have liver and renal disorder you can not use azithromycin and also you not use during pregnancy and lactation for children.
Side effect of azithromycin:
- Mild to moderate nausea
- Vomiting
- Abdominal pain
- Dyspepsia
- Flatulence
- Diarrhoea
- Cramping
- Angioedema
- Cholestatic jaundicem
- Dizziness
- Headache
- Vertigo
- Somnolence
- And transient elevations of liver enzyme values.
Drug Interactions:
If you take azithromycin with antacids containing aluminium and magnesium salts reduce rate of absorption. And increased risk of ergot toxicity.
Life threatening condition: Increased serum concentrations of digoxin and ciclosporin.
